Menurut RM BTS, "DNA" berkisar pada "ekspresi cinta muda yang penuh gairah" yang sangat berbeda dari rilisan grup itu sebelumnya, demikian seperti dikutip Billboard.
Secara lirik, lagu ini mengeksplorasi tema takdir dan cinta pada pandangan pertama, melalui kalimat-kalimat seperti: "Aku mengenalmu sejak pandangan pertama/ Seolah-olah kita telah saling memanggil/ DNA dalam nadiku memberitahuku/ Kau dialah yang selama ini saya cari."
Sementaraitu, secara komersial, "DNA" memulai debutnya di nomor dua di Gaon Digital Chart dan nomor satu di Billboard K-pop Hot 100 .
Sejak itu telah terjual lebih dari 2,5 juta kopi digital di Korea Selatan pada Februari 2019. Lagu ini memuncak di nomor 67 di Billboard Hot 100 AS dan di nomor 90 di UK Singles Chart, dan menjadi entri pertama grup di kedua tangga lagu tersebut.
